α-chymotrypsin is a serine protease that hydrolyzes peptide bonds at the carboxyl terminus of tyrosine, tryptophan, and phenylalanine residues.
Ca²⁺ serves as an activator and stabilizer of α-chymotrypsin. It is inhibited by diisopropyl fluorophosphate (DFP), phenylmethylsulfonyl fluoride (PMSF), N-p-tosyl-L-phenylalanine chloromethyl ketone (TPCK), chymotrypsin inhibitor, aprotinin, α₁-antitrypsin, α₂-macroglobulin, as well as 10 mM Cu²⁺ and Hg²⁺.
