RNase R is expressed in Escherichia coli with its sequence derived from the rnr gene of E. coli. It is a magnesium‑dependent 3'→5' exoribonuclease that efficiently degrades most linear RNA molecules, but does not digest lasso or circular RNA, double‑stranded RNA with a 3' overhang of fewer than 7 nucleotides, or some structurally complex RNA. This enzyme enriches circular RNA by degrading linear RNA in mixed RNA samples to increase the relative abundance of circular RNA. Its activity can be inactivated by heating at 65 °C for 20 minutes.
