DNase I is a non-specific endonuclease that degrades double-stranded DNA, single-stranded DNA and chromosomal DNA. It acts by hydrolyzing phosphodiester bonds to generate mononucleotides and oligonucleotides with 5'-phosphate and 3'-hydroxyl groups. The enzyme requires Mg²⁺ or Ca²⁺ for activity, with an optimal pH of approximately 7.8.
DNase I is obtained by recombinant expression and purification, and RNase activity is removed during the purification process.
