Vaccinia Capping Enzyme is expressed in Escherichia coli and consists of two subunits, D1 and D12. It possesses RNA triphosphatase, guanylyltransferase and guanine methyltransferase activities, enabling the attachment of a 7-methylguanosine cap structure (m7Gppp) to the 5′-end of RNA (m7Gppp5′N). This cap structure enhances mRNA stability, transport and translation efficiency, and is an essential modification for in vitro mRNA synthesis.
